LAS AF (Leica Application Suite Advanced Fluorescence) is a microscope control and imaging platform developed by Leica Microsystems CMS GmbH for advanced fluorescence and confocal microscopy. Designed for Leica TCS confocal and motorized widefield systems, it unifies hardware control, image acquisition, and analysis in a single workflow-oriented interface.
With LAS AF, researchers can configure multi-channel fluorescence experiments, acquire z‑stacks, time-lapse series, and multi-position datasets, and automate tasks such as tile scanning with stitching and hardware autofocus. On compatible systems, it supports spectral scanning and unmixing. The software saves data and rich metadata in Leica’s LIF format and includes tools for measurements, colocalization, 3D visualization, and basic quantification, along with export to common image and video formats for downstream analysis.
LAS AF integrates tightly with Leica lasers, detectors, stages, and environmental controls to promote reproducibility and ease of use. For offline review and sharing, LAS AF Lite provides a free viewer to open LIF files and extract images on computers that do not control the instrument. LAS AF has been succeeded by Leica’s LAS X platform on newer systems.
